First off I want to start by saying that the author should not be writing about technical aspects of which he is not knowledgable. If a dictionary source is used for a defintion, LEAVE IT ALONE- don't expound upon it unless you are a chemist with technical expertise. TLC is used when qualitative information is needed and is generally a simple, rapid test used to determine the presence/ absence of an analyte. The "plate of varying colors" may or may not occur depending upon the visualization reagents used in the method. HPLC, on the other hand, is a QUANTITATIVE method when primary standards are used and a P57 standard has become available. There would be no reason to run 2 qualitative tests; with HPLC it would be difficult to get a "false- negative" since these methods should be VALIDATED prior to use. The concept here is simple: if the sample in question has a peak which occurs at the same retention time as a primary standard it is regarded as positive for the analyte in question and can quantified. When you have competent people working in a lab using sound scientific principles you don't need to run lots of methods to determine the same thing. If consumers want hoodia gordonii then TLC would be sufficient. Companies that have products which claim to contain P57 should be able to provide the quantitative data (HPLC) to back up these claims. It appears that the author is paid by Alkemists Labs and may not be the most objective person to offer advice on the subject. I have evaluated this company's HPLC method and find that it could be vastly improved upon. I offer a warning about Hoodia: I have evaluated many products and have yet to find 1 that actually contains a quantifiable amount of P57. Consumers need to be aware that supplement companies can (and do) claim whatever they want on the products because there is no FDA regulation. So if someone is looking to lose weight DIET AND EXERCISE are still the best option.
